St. Clair was born and raised in Westfield, New Jersey where she attended Westfield High School. She is a graduate of Middlebury College.
St. Clair works is a regular sketch performer and improviser at the Upright Citizens Brigade Theatre in New York and Los Angeles, originally as a member of the improv group "Mother". St. Clair was a regular panelist on the VH1 series Best Week Ever, offering comedic insights on pop-culture and the news of the week. She is also a frequent guest on the popular comedy podcast Comedy Bang! Bang!, where she plays one of the show's regular characters, Marissa Wompler.
St. Clair and Lennon Parham are creators, stars, and executive producers of the NBC comedy series Best Friends Forever, which premiered on April 4, 2012. Her new comedy co-created with Parham, Playing House, will premiere on USA in early 2014.

St. Clair is married to playwright Dan O'Brien. Their first child, a daughter Isobel Kelly O'Brien, was born on October 1, 2013.
